My lab is in the process of designing an enrichment protocol for AHA-labeled proteins. The labelling efficency of our samples appears to be quite high (excellent signal to noise ratio with IHC and Western analysis), so the amount of labelled proteins should not be an issue. Has anyone had particulary good success with using alkyne agarose or the Dde cleaveable alkyne agarose to enrich AHA-labelled proteins. The Dde cleavable alkyne is a very attactive option, but I have not seen any papers with this. Would love to hear back from anyone that can offer advice.
