Que haces nena, porque no mandas un mail normal. NO SE como funciona esto y no tengo ni idea del CD25, pero aprovecho para mandarte un abrazo muy fuerte y desearte Felices Fiestas a vos y a toda la flia. Te queremos
I work mainly with clones 2A3 and BC96. Other clones probably work too. More important, I think, is to pick a bright fluorochrome. My favourite is APC, especially if you fix your samples after surface staining. BV421 and PE works fine too. FITC is more or less useless for CD25 on human PBMC in my opinion.
Also, titrate the CD25 antibody, often less than the recommended amount, gives a better separation between CD25+ and CD25neg cells.
I recommend a free antibody database available at labome.com. For antibodies to CD25 you can check the following link: https://www.labome.com/review/gene/human/CD25-antibody.html. eBioscience has several anti-human anti-CD285 abs suitable for flow cytometry: mouse monoclonal (BC96) (Patel et al, J Immunother Cancer. 2016), mouse monoclonal (RPA-T4), mouse monoclonal (CD25-4E3). Also, Biolegend offers mouse monoclonal (BC96) ab. Invitrogen offers mouse monoclonal (CD25-3G10) (Xu et al, Immunol Invest. 2012). BD Biosciences has mouse monoclonal (M-A251) anti-CD25 (Cook et al, Oncoimmunology. 2015). Also, Beckman Coulter provides mouse monoclonal (B1.49.9) anti-CD25 (ampion et al, J Exp Med. 2014). You can check the description of these antibodies and the references following the provided links.
