I would like to measure concentration of BCNU anticancer drug in PBS solution using UV. For some reason, my peak is really broad (peak almost near 340nm) with band width over 200nm and it has a right shoulder. I don't filter my sample because I have limitations with volume. Also my sample would have PLGA (poly lactic glycolic acid) along with BCNU. Does anyone have suggestions for getting a sharper peak for BCNU?
Hi, What functional group(s) in BCNU that give rise to absorption max at 340nm ? PLGA also does absorbed ~ 210nm. To sharper the peak narrow the bandwidth. But I don't this will help you as your BCNU UV is masked by the absorption of PLGA or other impurity.
In liquid phase it is normal that the absorption spectra present broad peaks. In your case the first step to follow is to try to separate BCNU from PLGA.
Do you use solvents? If you are using solvents you can fine tune the spectra changing the solvents.
how about the puriy BCNU, if there is broad peak in purity BCNU, may be you can find other suitable method to quantilify the concent in pbs
