Hi, together with my research group we are attempting to optimize a procedure for extracting bacterial DNA from a contaminated textile matrix (used FFP2 mask). Currently, using a DNA extraction kit from blood we have obtained minimal yield (almost zero) and high levels of DNA contamination. Are you aware of any similar literature experiences or procedures that can optimize the yield and quality of the extract? Thank you

I think if you treat it like tissue, put in liquid nitrogen, homogenize and complete the steps using tissue extraction kit could give better result.
DNA concentration will be low anyway as the bacterial or viral content in the matrix will be almost negligible.
Try using platinum polymerase to increase sensitivity of the PCR.
