I’m doing pei max transfection in platinum E cell lines to collect retrovirus supernatant but my transduction is very low. if I add sodium butyrate as booster for enhancing transfection and increase viral titers at what time need to add this and how to remove this from Supernatant?
because this is cytotoxic to T cells during transduction.

You typically want to add sodium butyrate 6–8 hours post-transfection and incubate for 8–12 hours. After that, remove the medium and replace it with fresh medium. This step helps eliminate residual sodium butyrate while allowing sufficient time for it to exert its effect as a histone deacetylase inhibitor, enhancing viral production.
Additionally, consider adding polybrene (4–8 µg/mL) during transduction to further improve efficiency by facilitating viral entry into target cells. However, unfortunately, these steps probably need to be empirically optimized because they're a lot of variables at play.... cell type, concentrations, etc
