In a phagemid system after infection phagemid containing bacteria with helper phage two kinds of phages are produced and extruded: those presenting protein coded by phagemid in fusion with coat protein and wild-type. Has anyone know if wild-type will be extruded first or at the same time? I have an idea how to check it, but maybe it's not neccesary. I work with M13K07 phage and pSEX81 phagemid.
yes you will have a mixed population of phages, some will be the M13KO7 and some will be the pSEX81 phagemid. I can't predict the precise rations but there will be lots of each.
You can easily check by infecting a host and plating dilutions on kan plates for K07 and on Amp for pSEX and counting the plating efficiency.
