I've working on trouble shooting qPCR within my lab on some porcine vaginal samples. I converted the RNA to cDNA and selected two house-keeping genes (B-actin and GAPDH), based on previously published work that utilized these two as house keeping genes. The B-actin shows up in the target tissue, but we have been unable to make GAPDH appear in acceptable levels. I will proceed in using the B-actin as one of my house keeping genes, but would appreciate any suggestions into alternatives to GAPDH as a housekeeping gene.
