I'm trying to work through the protocols for TUNEL staining using In situ Cell Death Detection Kit (Roche) to detect apoptosis in adherent cancerous cells, and the recipe of the permeabilization solution keeps bugging me. Are there any tips such as the best permeabilization buffer or time and temperature of incubation?

You can try the Fixation/Permeabilization solution kit from BD, proceeding according to data sheet. Always works for me.

Aline Zaparte Thank you. But I don't access this kit, so do you know which concentration of Triton X 100 needed for more effective permeabilization?
