I want to identify/sort X- and Y- bearing spermatozoa on frozen-thawed bull sperm samples using Hoechst staining by flow cytometry. Should I use raw semen before flow cytometric analysis or can I also use my frozen-thawed samples too?

For this purpose I can guess there should be no difference. As long as sperms are alive.
Flow cytometry always works much better in live/freshly extracted cells.
