The spleen is highly affected by systemic infections and consequently suffers from several disorders and problems. But are there any biomarkers or tests through which we estimate the functionality of the spleen?

You can dose the chitriosidase. It is a marker of macrophage activation. The spleen is extremely rich in macrophages. Dysfunction of the spleen can be related to massive macrophage activation. Chitotriosidase is a surrogate to test the activity of macrophages. In clinic it is used in many disease, including sarcoidosis and Gaucher disease
