My plan is to do free-floating sections for IHC of tyrosine hydroxylase. My questions are: 1) should I dehydrate the sections before mounting them onto slides? 2) I think I need I do a counterstain after IHC. What’s a good counterstain for neuron labeling after the IHC? Nissl-staining? 3) Also, can some please suggest a good mounting medium for DAB-based IHC of brain sections which have a high lipid content?

In order to mount your brain sections onto slides, you should use the following protocol: 1) water 5 - 10 min 2) EtOH 50% 5 - 10 min 3) EtOH 70% 5 -10 min 4) EtOH 100% 5 - 10 min 5) Xylene 5 -10 min 6) Xylene 5 - 10 min 7) Eukitt.

PS. You should avoid to dry your brain sections before mounting each of them with Eukitt....so, for this reason, keep your brain sections in xylene 2 until the step with Eukitt!
