Has anyone analyzed the stability of protoplasts at ph 8.0-8.5?

No I haven't, but even if they are stable I wonder how many pH-dependent PM localized proteins (channels, pumps and many others) would be affected by such a high pH (high compared to the normal cell wall pH).... you should probably take this into account when performing subsequent experiments.... good luck.
