Dear All, I am new to this Normal Human Astrocyte (LONZA). They are primary culture. The first vial of cryopreserved cells that I revived seems ok, however, another does not. I noticed a big clump of "substance" on top of the astrocytes, and there are also round, clear "bubble" on the cells. May I know are they stressed?

I wanna answer you from my experience on adherent cells (normal or cancer human, murine cells) but not on astrocytes.
In general, the cells seem to be not so bad if you passed them recently. These photos after how long time incubation in the culture medium? because we see the philopods that responsible of attachment to the surface and they do not have stress form or at least the stress form that I used to see with weak adherence and approximate round shape without long and multi philopods, unstable movement of the cell all the time (via cell live image). I see as well a cluster of cells in the middle. I saw that in some cell lines and when the flask had high confluence but I guess that is not the case here, only if they become from another passage where they were not very well disassociated after being incubated with the enzyme. The different type of cells is normal in primary cells culture. You need to sort out the cells to make sure what are exactly the cells you isolated or whether they are the requested cells or not.
