I am cutting fish larvae after their embedding in Technovit 7100. I usually use methylene blue/azure II/basic fuchsin in the staining phase. However, I noticed that for some tissue structures it is not an appropriate staining, so I would like to try with H/E. Is there anyone with experience that would like to share with me infos/protocols/suggestions?

Hello Barbara, I found a staining recipe which is recommanded by Kulzer. I recieved it years agoe. The procedudre:
staining of the sections in Gill's hematoxylene for 15 min, running tap water for 10 min, rinsing in destilled water, staining in Eosin for 2 -5 min, dehydration in 96% and 100% ETOH (several dipps each), clearing in xylene and mounting in DPX or Eukitt.
The solutions: a) Gill's Hematoxylene: Hematoxylene (C.I. 75290) 6 g, Na-Iodat 0,6 g, Al-Sulfat 52,8 g, destilled water 690 ml, Ethyleneglycol 250 ml, aceitic acid 60 ml. b) Eosine: Eosin-Y- (alcoholic) (C.I. 45380) 0,5 g, 96% ETOH 100 ml, acetic acid 2 drops.
If you like to decolorize the methacrylate-matrix dip the slide into a solution of 0.5% HCl in 70% ETOH just for several times. When the matrix is clear put the slides in tap water and continue the staining process. Good luck!
