Currently, I am trying to synthesize silver nanoparticles from Enterobacter aeroegenes. After I inoculate the 100 ml nutrient broth with the bacteria and incubate it at 30 degree. After 24, 48, and 72 incubation I take the biomass then I centerfudge them for 10 minutes at 10000 rpm, then I take 50 ml of the supernatant and add to it a 50 ml of 1mM siver nitrate solution then I leave the flask in dark at room temperature. But I got no color change eventually! Any help or maybe something else to do?

If the color of the nutrient broth is yellow, the formation of Ag Nps will be hardly detectable by visually. This is due to the light yellow color of Ag Nps. You should check the sample by TEM. If the formation of Ag Nps cannot be observed, you should performed the synthesis at elevated temperature.

I think you have to check your sample by UV visible spectrometer, because AgNPs at low concentration shows yellow to brown color. If you get any significant peak in UV spectra that will be your confirmation for synthesis.
And remember one more thing, what incubation time you are using that also depends, try to incubate for long time compare to your present exposed time.

Sometimes there is a possibility that it can produce small nanoparticles, hence you wont find much color changes. So check the solution under UV and record UV vis spectrum. if you find any changes in the peak position there is more possibility for AgNPs.
compare the extra cellular solution UV and AgNPs solution UV spectrum.
Observe both of them under UV light (365 nm) if it is fluorescent you can identify easily.

bacteria like ag259 can be used for silver NPs synthesis. can produce NPs in good amount.
