We want to inhibit the binding of BDNF and NT-4 to its receptor. So we are looking for some peptide inhibitor or antibody that can binds to these proteins in their binding sites. Any suggestion? Thank you.

Hi Erica, I run a neurotrophin lab as well as act as an advisor to Biosensis (biosensis.com). They have some excellent blocking antibodies to all the neurotrophins that are well characterized in terms of cross-reactivity, dose, etc. The best characterized are sheep antibodies. Antibody #9 is a great BDNF neutralizing antibody developed by Yves Barde. It is very affordable and available through the Hybridoma bank at the Univ of Iowa (http://dshb.biology.uiowa.edu/brain-derived-neurotrophic-factor). They do ship internationally. Alternatively, K252a will block Trk signaling. Although it is a dirty drug, it is an easy tool to test and support your hypothesis. Also the dominant negative T1 TrkB has been use to shut down signaling if you can transfect the construct in your system. Good luck!
Number 9 does not work well in traditional westerns. Its companion, #1 works better as was demonstrated in the Kolbeck J. Neurochem., Val. 72, No. 5, 1999 paper. IP will work with #9 though if this helps.
