You can isolate them from peripheral blood, but they will die after a few days in culture, even if you add IL-5 to improve their survival (these cells do not proliferate).

I was used to culture them in RPMI1640 +1%antibiotic-antimycotic +1% L-glutamine (200 mM stock). Eosinophils are usually just plated in multiple well plates (not tubes) without agitation. Incubate at 37°/ 5% CO2 in a humidified incubator as usual.
