I want to isolate fish brain specific zones like telencephalon, cerebelum, opticum tectum. I am encoutering with the problem of hardness of tissue because in the moment we try to take fish brain from the skull the tissue is very soft and it is very tough to collect the whole tissue to further separate specific zones. Anyone who can give me some tips to carry out this sucessfully?

I havenot work in fish brain but your problem might be solved by using vinegar, 5% acetic acid. What acid does it hardens the tissue thus you can pick or dissect into separate parts. This technique works quiet well when I was working to separate the gonads of small fish (3- 10g) from its body.
