I have a drug-metal complex which is soluble in DMSO-Water only. And I need to look for its stability in-vitro in cellular medium. I am planning to do a dialysis where the release of drug in buffer can be studied over time.
What are the molecular weights of the drug and of the drug-metal complex? 300 Da is a very small cutoff for dialysis tubing, but it may be possible to use a more readily available larger cutoff membrane if the drug-metal complex is much larger.
Are there spectroscopic methods that can be applied to the problem instead of dialysis? For example, is there a difference in the absorption, fluorescence, EPR, or NMR spectrum of the free drug and the drug-metal complex? Can you test for the presence of the free metal instead of the free drug using a metal-dependent fluorophore? Can you use HPLC to analyze the mixture for the free and complexed drug?
