I need to analyze samples to measure startch contents from fruit. I am having trouble to make the standard for starch. In the protocol, starch assay part and Glucose assay part is not clear enough. Can anybody help me if has done earlier. I have attached the file for your convenience. Thanks in advance.
We mostly use Megazyme kit for starch analysis of cereal and legumes starches.
I analysed starch by the polarimetric method. but I have determined glucose in juices by the GOD POD method Spanish kit. this last method is spectrophotometric. it is recommended to prepare diffrent solutions of solid starch to be heated and used as rferences. the final DO at 340nm should not exceded 0.9 to be reliable
This kit is for the quantitative, enzymatic determination of native starch in food and other materials. The hydrolysis of starch to glucose is catalyzed by amyloglucosidase. Glucose is phosphorylated by adenosine triphosphate (ATP) in the reaction catalyzed by hexokinase. Glucose-6-phosphate (G6P) is then oxidized to 6-phosphogluconate in the presence of nicotinamide adenine dinucleotide (NAD) in a reaction catalyzed by glucose-6-phosphate dehydrogenase (G6PDH). During this oxidation, an equimolar amount of NAD is reduced to NADH. The increase in absorbance at 340 nm is directly proportional to the glucose concentration.
