I'm trying to test the efficacy of a recombinant vaccine for botulinum in bovines. I would like to test the antibody responce by using a L+ test. I've read the european pharmacopoeia instruction but I'm a little bit confused. Anybody can help me? do you have other litterature?

L+/ based neutralization method for titration of antitoxin date back to more than 100 years, described by the Paul Ehrlich and is the Gold Standard method for determining potency of antitoxins. L+/ is defined as minimum amount of toxin when combined with 1 Unit of antitoxin will cause death of 100% animals within a specified period, for example within 4 days for tetanus antitoxin method. There have been excellent reviews and articles on L+/ based potency methods. The choice of specific method (toxin dose level) depends upon the level of antitoxin in test sample. It seems that you are determining antitoxin in animals injected with vaccines and you would need low levels of toxin. I have published on titration of tetanus and diphtheria antitoxins using L+/ based neutralization method at various levels of toxins. You may refer to the attached papers.
