Hi,

I have been trying to record Parvalbumin interneurons in mouse brain slices. I tag the PV cells with AAV. However, most labeled PV cells are not on the surface of the slices. I'm having difficulty in patching cells deep in the slice. The successful rate is very low. I think the most difficulty is that it is hard to tell if the tip is touching the target cell or not. Most time I was not able to form giga seal. I'm wondering if any one get give me any advice on this issue? Many thanks!
