I have problem with the mEPCs recently. When seeded the bone-marrow-derived mononuclear cell in fibronectin-coated 6-wells plate with complete EGM2 medium(lonza) supplemented with 15%FBS and 1%antibiotics. At day 4, non-adherent cells were removed and medium was changed every other day. However, beween day 7 to 28, most of the cells seems to show the characterization of mesenchymal stem cells, which is not in accordance with related articles. Do you have any suggestion about this?
I would suggest that you first remove majority of the monocytes/MSCs from the fraction by plating on uncoated plates for ~3 hours, remove the unmatched fraction and plate on fibronectin-coated plates.
I used to work with mouse endothelial progenitor cells derived from bone marrow and one key point for our laboratory was to change medium everyday in the first 7 days of cultivation.
