Hi all, we're currently working on T cells and considering getting a nucleofector device for highly efficient transfection yield of Jurkat T cells. Does anyone have experience about this device? I've looked up the market info and found 2 most popular devices: Amaxa and Neon. I used to tried Amaxa one before from another lab, but the efficiency is not good, only about 20% successfully transfected. I'm not sure if it's about the device or poor sample preparation. I'm hoping to hear some experiences and choices or yours.

We've used older electroporation devices, but the ones you listed have been used in research and should probably work, as long as you have proper electroporation buffers for Jurkat (https://altogen.com/product/jurkat-electroporation-kit-t-cell-leukemia-tib-152/). Make sure you have the protocol right, as even though the device is important, knowing the parameters for electroporation will be more important for overall efficiency.

As you know, often times it is very difficult to achieve high delivery efficiency when using an electroporation approach in T cells. May I ask if you have tried Self-Deliverable RNA silencing FANA Molecules to achieve high delivery efficiency in T cells?
If not, I encourage you to review the attached FANA delivery protocol without the use of any transfection reagents.
