My lab is currently looking at creating a custom antibody. There aren't a lot of customer reviews on the websites for these things, so what are the companies that you all prefer to synthesize your custom antibodies? Price & quality are my biggest concerns. Polyclonal is fine.
Our antigen is a human/rat protein. I'm just looking for your personal thoughts about and experiences with custom antibody providers so that we can use the company that seems to be most well-reviewed by the scientific community.

I've done that during my thesis about 10 yrs ago, but since polyclonal is fine for you affinity purification of antisera might not have changed much since.
We ordered rabbit antisera made with GST fusion proteins at Cocalico Biologicals, 2 per antigen, for about $400 each. The delivery was a test bleed followed by 5 bleeds of about 15 ml of serum and a bigger final bleed to use so or affinity purify. The endogenous protein was not very abundant, never detected it in mass spec, we affinity purified the sera with the same antigen in a Maltose Binding Protein (MBP) fusion immobilized on a Actigel ALD column.
Once glycerol stocks for fresh Ag-MBP fusion production were there, it was about 1 wk of work from the 15 ml antisera to the 1,5 affinity-purified concentrated antibody. I did 3 maybe 4 purifications over 5 years and had way enough Ab.
It worked very well where all our tests of commercial antibodies were unsatisfactory. However don't be afraid to test incubation conditions for the antibody, milk type, BSA, buffer and temperatures made a huge difference for me, but this is also true for antibodies other than custom made.
