I used PCR for detection of P. vivax CSP, multiple size bands were detected. i found many researches about the detection but mostly using ELISA , what am questioning about is the detection of size length which is less than 500 bp while it was never detected in humans. what factors could be attributed to this variability?
What primers do you use? CSP contains a region of tandem repeats, it is important to select primers outside the repeat region.
The PO1 reference strains contains 17 repeats of 27 bp each. Thus the size of the repeats is 17x27= 459 bp, thus even if you select primers just flanking the repeats, the size of the PCR product will be around 500 bp. Your samples might contain fewer repeats, but the fact that you see several bands might indicate that at least one of your primers is located in the repeats region and binds several of them.
Due to the genetic diversity (i.e. different number of repeats), you should expect to see bands of different sizes in different samples, and more than one band in case of multiple clone infections.
