I am trying to isolate lymphocytes from mouse small intestine for primary culture them. However, I have a large amount of epithelial cells, and I can't get rid of them. Any suggestions?
Have you ever tried running a ficoll/hypaque separation after collecting Peyer's patches? I'd go for that and a negative selection may help you as well, by using magnetic beads conjugated with an anti-EpCAM Ab. You can find attached a nice chapter explaining different approaches to deal with what you're asking. Good job!
We have already tried ficoll, however we still have a lot of epithelial cells. Unfortunately, we do not have time for magnetic beads at this moment, but I believe this chapter will help us a lot. Thanks
