We use for mordant thymol (3-4 crystals at 100ml of the filtrated gelatin solution) You will have bacterial contamination without mordant. It will give problems with adhesion of sections on slides a artifacts after staining
You may also use not gelatin but albumin-glicerol by Mayer: white of the egg mixed with pure glycerol at volume 1:1 + 2-3 crystals of thymol and filtrate through 2-3 layers of filtrate paper (better do it in refrigerator 4C, because it`s very slow!!). On dry slide drop little drop of this solution and rub it steady
A week ago I prepared Mayer´s albumin-glicerol. Today I used it for the first time and many of the sections were dropped off the slide during the staining process (Hematoxyline-eosine). I did not filter it with paper (the protocol I followed did not say a word about that).
I am using polyster wax instead of parafin. It is my very first experience with it and I am not sure if the sections loses were due to not filtering the albumin-glicerol or it is due to the wax I am using. Any piece of advice?
