I have measured the total cholesterol and triglyceride from 0.5 g of mice feces. First I extraction total lipid with Folch et al., J Biol Chem 1957, 226, 497 procedure, http://www.cyberlipid.org/extract/extr0005.htm and then dry it down with Nitrogen gas. When the lipid is completely dry I re-suspended it in 100% Ethanol, from here I used 5 micro-liter for the total cholesterol measurement with the total cholesterol kitand standard from Sekisui-biopacific Inc. Hope that helps, good luck.
Perform a bligh & dyer extraction, resolve the extracted lipids in H2O triton 2% , then a standard available cholesterol kit could be used ( colorimetric or fluorimetric )
