I am trying to block some ion channels putting antibodies in the ICF (intracellular fluid) using whole cell patch clamp.
The antibody stock's concentration is 0.85 mg/ml and I am using it 1:100 (as I saw in this paper https://www.ncbi.nlm.nih.gov/pubmed/21576272)
I saw an effect but it's kind of puzzling (very strong and fast, approx. 10mins after membrane rupture) so I was thinking to use it more diluted, maybe 1:400 or even more.
What concentration are you usually using?
Have you ever tried different dilutions?
