Is there any difference if you do it with L broth vs MHII broth? I know MHII has starch, which neutralizes toxins and has better diffusion on agar, but what if you use L broth instead?
Muller hinton agar or broth are standard culture for antibiotic resistance testing . The luria or l broth is standard media for molecular biology study , such as plasmid or DNA conservation , because is more specific and differential media than muller hinton .
Mueller Hinton type 2 (or MH2 or MHII) is a bacterial culture medium with an adjusted concentration of cations. This medium should be the medium to use when carrying out an antibiogram or an antibiotic sensitivity test (liquid or solid phase).
It is important that researchers always use the right one and the same medium when they carry out the same type of experiment... Why? because it is important to follow the recommendations and above all, it allows to compare the results between them. Finally, it must be understood that two different bacterial culture media have different compositions, and therefore these different compositions (i.e. in substrate) will necessarily have an impact on the growth of bacteria... there is therefore the risk to introduce a bias, and to compare things that in the end are not comparable.
