The cell monolayer does not appear confluent after three days of incubation. Numerous gaps between cells are evident, which is atypical for fibroblast cultures under standard growth conditions. This lack of confluency could be attributed to:

A low initial seeding density, or

A potential cytotoxic effect exerted by the 5% extract.

3. Cellular Debris:

The presence of dark particles and granules within the culture may represent cellular debris, suggesting the occurrence of cytolysis or compromised cell membrane integrity.

4. Interpretation:

The morphological changes, reduced confluency, and presence of debris collectively suggest that the 5% extract may have negatively impacted cell viability. It is plausible that the extract inhibited fibroblast proliferation or induced cytotoxic effects. Given the observed condition of the culture, it appears that the cells did not maintain normal growth and morphology throughout the three-day incubation period.
