I had a general question in regards to purification of vector and insert after a restriction digest. Would it be possible to use an AmpureXP magnetic bead clean-up (using Qubit for dsDNA quantification afterwards, and BA to view DNA profile) after a restriction digest, instead of gel purification? I'm a little new to this, and might need to optimize a protocol for yeast insert cloning.
Thank you, Mathew. I had my suspicions of whether this would work because I know that AmpureXP clean-ups tend to be less efficient for higher fragment sizes. Though in my mind, I thought about doing a 1.8-2X clean-up to get all of my fragments, but maybe I could try, and run in parallel with the gel purification, and run another gel to see if they look comparable.
