I need quantify the content of amino acid in the supernatants from a fermentation experiments with Corynebacterium Glutamicum.

For that purpose, I plan to employ the ninhydrin method as it appears to be a common method for amino acid quantification of this type of sample.

In the literature, there are different methods for preparing the ninhydrin solution and I am a bit confused which one will be best to use.

In the paper "Optimization of Fermentation Media for Enhanced Amino Acids Production by Bacteria Isolated from Natural Sources", ninhydrin solution is prepared by mixing phosphoric acid with glacial acetic acid and ninhydrin?

Before I proceed in the lab, I would like to ask if anybody can share there experiences with this method and recommend me a recipe for the preparation of the ninhydrin solution.
