Test tubes containing 5 mL of presterilized medium of composition: 1 g of yeast extract, 0.4 g of ammonium sulfate, and 0.15 g of potassium dihydrogen phosphate (pH 8.0) were inoculated with organisms from individual colonies of the isolates and kept one test tube as control.
Does the pH 8.0 mean the pH of the medium as a whole is 8.0 or the pH of the substance (in this case, potassium dihydrogen phosphate) 8.0? Also, are the pH values natural or do I have to add a specific amount of acid or base to achieve the needed pH?
