In Alpha glucosidase assay does PNPG (from SRL) degrade readily if kept at room temperature for about 2-3h?

It depends on the pH of your buffer. If i remember correctly, at lower pH PNP-substrates will degrade quicker (you can easily check how much is degraded by OD measurement).
