I wondering which of these labels is better for standard epifluorescence. I have been happily using Alexa546, but I recently got some fading problems, and someone recommended me to try Alexa555 which he said is much better.
I then asked the Life Tech support for advice but don't really quite understand what they mean. Apparently A546 has much bigger quantum yield and lifetime (see https://de-de.invitrogen.com/site/de/de/home/References/Molecular-Probes-The-Handbook/tables/Fluorescence-quantum-yields-and-lifetimes-for-Alexa-Fluor-dyes.html), while A555 has somewhat bigger extinction coefficient (https://de-de.invitrogen.com/site/de/de/home/References/Molecular-Probes-The-Handbook/Technical-Notes-and-Product-Highlights/The-Alexa-Fluor-Dye-Series.html).
I apologize for my ignorance but I don't know what do these mean in practice. If I want a bright signal that doesn't fade easily, which one should I choose?
