I have an urgent question. For growing aerobic microorganisms, I prepared conical flasks which covered with cotton at top. After 3 months I decided to measure the amount of produced CO2 by a gas analyzer. Therefore, I designed a rubber with a single hose inlet, and tight the coming hose to prohibit the gas existing. Now, I wondered by doing this I change the condition to anerobic.
Please note that every 4 days after measuring the captured CO2, I open the cap for 1 min and then close it as I explained.
I urgently need your advice in this matte, please.
So what was your question. did you have a growth or CO2. if there is then it may not be anaerobic completely.

Your questioin s not clear, please specify your problems: do you want an anerobic culture system for your aerobic organism or you're just doubtful that your culture system might become anerobic, due to a tight cotton plug or for closing the exit of an inserted rubber tube into the cotton plug; make the purpose of this experiment clear. One point to mention here: "be sure that just a tight cotton plug or by closing the exit of an inserted tube can not make a culture anaerobic. So, I doubt the produced CO2 gas already escaped your system. 3 months is a long time for an aerobic organism, so I think the organism has already grew out, and the CO2 escaped. Please check any indication of growth and /or presence of substrate (C source) in the culture medium. is already It might be already consumed up. Good luck.

The answer on your question depends on "design" of your conical flask.
1) WHat is total volume and what is volume of gas & liquid phase - you can calculate how much O2 is in the flask.
2) What is your medium look like: is it rich organic medium or another type of medium.
3) what is the growth substrate - if you cultivate your bacteria on certain amount of substrate + you measure produced CO2. You can calculate how much oxygen was reduced and then you may estimate how much oxygen left in your flask.
