To form adventisous root, plants require auxin (endogenous/exogenous) and high humidity (RH). Since solanaceae family have high rofile of endogenous auxin and there are enough humidity in vitro, root initiation occur in stem nods of the plant.
Shoots regenerated in vitro can be transferred to medium without auxins. Roots formed are not aerial. Our experience from petunia and tobacco tissue culture.
As you didn't mention about the current in vitro culture condition as well as the previous condition, I suggest you try to use the aerated vessel instead of airtight vessel in order to reduce relative humidity inside the vessel. If auxin was used in the previous cultures, I also suggest you add some activated charcoal into the culture medium in order to absorb accumulated auxin which can stimulate the root formation thereafter.
