I am working on adhering insect chironomid larval cells to polycarbonate membrane but the cells seem to get lost after washing with PBS. I have been able to adhere these cells to glass by using collagen and BAM(biocompatible PEG anchor) which adheres very well, but this has not worked for the polycarbonate membrane. Any advice on what to do? Thanks
Some cells lose the ability to attach to culture plates while the passage numbers goes up.
Overnight coating of the plates with 1-2% sterilized gelatin solution (in MilliQ water) can help increase the attachment rate. Also, u may try different cell seeding densities in such plates. If the cells are over crowding beyond the expectations, reducing the serum conc. can help (if you are using serum for the plates).
