Because spinal cord samples contain high levels of lipids, I was told that adding SDS to the sample prior to dilution and BCA assay will allow for better/more precise readings.
1. Is there any literature to support this?
2. What is an optimal amount to add (i.e. to a max of 5%)?
3. Are there any protocols that can be referred to?

1. I came across this article supporting that SDS do help to extract protein in your case; http://www.ncbi.nlm.nih.gov/books/NBK21589/
2. Not all BCA assay kit can tolerate SDS. For example BCA protein assay kit by Thermo can tolerate 5%, while 2-D Quant kit can only tolerate up to 2%. So you have to check which kit you are using.
3. As for specific protocol, i did not have them in hand. But I guess you can find it over the PubMed.
