Hello folks,

I found that my E.coli BL21 DE3 star cultures grew to O.D. 1.2 before I could induce with 0.5 mM IPTG. I normally induce at O.D. between 0.4 to 0.8 when they are in exponential phase at 37 degC, and drop them down to 30 degC to allow expression for 5-7 hours (O.D. 1.6 - 2). The plasmid carries Kan resistance.

If I were to add more LB media to drop the O.D., would the cells be 'rescued' and still produce as much protein? The protein is His-GST tagged and soluble.

Please let me know if you need me to provide any more details.

Cheers,

Pramod
