In the test to check the ability of lactic acid bacteria to growth under bile salts (Bile salt N3) where control sample (Bacteria + MRS broth) and treat sample (MRS+ bile 0.5% + bacteria) how to deal with the blank (MRS + 0.5% bile) absorbance for the treat sample. The OD of the blank for treat sample calibration shown by the spectrophotometer is saying "Too high absorbance values " ( the solution is opaque). In this case the treat samples can`t be measured at initial time before further incubation to test the survival rate of the bacterial strains. 0.5% bile was added to the MRS broth after being filtered through 0.22 μm membrane filter.
