How do you convince yourself that your drug candidate is effective in the intended indication area?

there is a section in the first overview depicting the steps employed to strengthen your idea as target identification and target validation both based upon genetic or molecular level info.
an obviously rather filosophical question, but in my experience: does the mechanism makes sense ? and secondly, is there an dose or exposure/effect.
Other aspects relate to feasibility, how do you get the drug in the 'system', and is it safe (eg QTc ex vivo assesment, drug-drug interactions, potential PGx, are metabolites active). It's not 'by accident' that the pipeline of products start with a lot of compounds to end with some 'survivors'
