Recently, i tansfered plasmid into mammalian cells（293T），but，i did not get a satisfied result. I have try diffenent voltage, such as 160V,170V,180V,190V,200V,230V,250V. When the votage is 180V, the expression of plasmid is higher than other conditions.By the way, i used a 4mm cuvette, the electric capacity is 950uF. I transfered about 4ug plasmid into one million 293T cells.So,i want to know how i can increase the expression of plasmid and stablized the expression of protein. Thank you!

Is there a reason why you chose electroporation rather than other methods? The reason I ask is because there are alternative methods that can highly transfect 293T cells (Calcium phosphate and lipofectamine). So if you keep having trouble with electroporation I would give the other methods a go.

I fully agree with Mahmood. Why choosing electroporation for such standard easy-to-transfect cells as 293T cells? All available chemical reagents or Ca-P transfection will give you 100% satisfaction without damaging your cells. And without problem of adjustment.
