I would like to evaluate myeloid (monocyte/macrophage/Dc) content of my tissue lysates by q-PCR. I did it the same with T cells by quantifying rearranged TCR DNA which is specific to the T lymphocyte lineage. It s easy and works great, and it s much more convenient than using RNA. I would like to find the same kind of assay for myeloids, basically quantifying a DNA which is specific to this lineage. I know I can use RT q PCR and check for myeloids specific expressed RNAs, but it will be way more complicated to correlate witch a number a cell, since a cell can express 2 RNAs or 10000 RNAs of the same gene. Any idea if it s realistic? I m scratching my head around MHC II , but it s not expressed only in myeloids. Any suggestion is more than welcome
