I was wondering if it is possible to detect a modified nanoparticle (5nm diameter) in the binding site of my protein using X-ray crystallography. The protein I want to co-crystallise with this "ligand" is around 70/80 KDa. I am not sure if x-ray can help me detect a huge ligand that is almost as big as the protein and I am also wondering about the problems I may face during co-crystallisation. What do you think?
