I co culturing PBMC and MSC cells. PBMC become so adhesive efter 72 hour incubation. IF I used PBS with 2mM EDTA, can Mesenchymal cells detach too? I dont want mesenchymal stem cells detach! has anyone tips about this?!
Chances are that they will detach. Mesenchymal cells in my experience trypsinize quickly, so I'm sure EDTA will have the same effect, as it will chelate ions necessary for integrins to work. Since mesenchymal cells tend to move around a lot, not sedentary like epithelial cells, they aren't as "sticky". You might be hard pressed to get the PBMCs out and not the mesenchymal cells.
However, if you wanted to at least separate them, you could do a differential trypsinization, where you add it for 3-5 minutes and the those cells, leaving the PBMCs behind. Then you can just pass the MSCs. Good luck!
