293T cells are not spreading to form monolayer. What is the reason and Solution to this?

When I infected 293T cells and MS7 cells with Lentivirus they lost their monolayer property and started growing on top of each other. They are under puromycin selection.

My labmate did Crisper- cas Knockdown, she also experienced the same thing.

Please seggest what can be done to solve this.

Looks like inhibition of adhesion. This event is independed of cell type in your case, so try to optimize the infection protocol. Try either to reduce lentivirus titer or remove the virus from the cells right after incubation period or sooner, than it is prescripted in the protocol.
