16s rRNA sequencing for bifidobacteria spp level identification?

Dear all please share the methodology for identification of specie level identification of bifidobacteria. I use gram staining, catalase, oxidase, indole, F6PPK for genus level identification but don't know how to identify specie level.

Is API kit reliable for bifidobacteria?
